Can I dispute a large charge from a reoccurring service?

Usual charge is $18/30 a month, suddenly got hit with a $525 charge. Can’t dispute it until it goes through, but am I able to dispute it? Unable to reach merchant due to them being a small business and off (conveniently) today.
